چکیده

Several powerful mechanisms exist to prevent self-reactive B cells from maturing to secrete potentially destructive autoantibodies. These mechanisms act in addition to the removal of self-reactive helper T cells.

Autoreactive anti-DNA transgenic B cells in lupus-prone New Zealand black/New Zealand white mice show near perfect L chain allelic exclusion.

Recent work on B cell tolerance and autoimmunity has suggested the L chain allelic inclusion is a property of autoreactive B cells and is closely linked to receptor editing. Allelic inclusion could rescue autoreactive B cells from clonal deletion by reducing their effective BCR surface density. We have investigated this phenomenon in anti-DNA producing hybridomas, derived from different strains...

B cell tolerance checkpoints that restrict pathways of antigen-driven differentiation.

Autoreactive B cells can be regulated by deletion, receptor editing, or anergy. Rheumatoid factor (RF)-expressing B lymphocytes in normal mice are not controlled by these mechanisms, but they do not secrete autoantibody and were presumed to ignore self-Ag. Surprisingly, we now find that these B cells are not quiescent, but instead are constitutively and specifically activated by self-Ag. In BAL...

Autoimmunity and the pathogenesis of myocarditis.

T he ability to distinguish between self versus nonself is the most salient feature of the immune system. It is of vital importance for the immune system to mount vigorous humoral and cellular responses to foreign antigens. On the other hand, it is equally important for the immune system to be tolerant against self-antigens. Tolerance can be maintained either by physical elimination (clonal del...

Self-reactive antibodies (natural autoantibodies) in healthy individuals.

Antibodies that are present in the serum of healthy individuals in the absence of deliberate immunization with any antigen, are refered to as natural antibodies. A vast majority of natural antibodies react with one or more self antigens and are termed as natural autoantibodies. The importance of natural autoantibodies in immune regulation has long been neglected, since tolerance to self was tho...

Induction of Mixed Chimerism for Reversal of Autoimmunity in Type 1 Diabetes

Type 1 diabetes (T1D) results from autoimmune attack of insulin-producing pancreatic islet β cells. T1D autoimmunity is associated with particular MHC or HLA types in mouse or humans. T1D autoimmunity arises from defects in both central negative selection and peripheral regulation of autoreactive T cells as well as intrinsic defects of B cells. Current therapies that target at improving periphe...
